it did look like cotton I think. The spot is quite small less than a millimetre. One of the debauwis is lying in the corner of the tank, barely moving. Is there anything I can do for him ?

I dont think you appreciate How high the nitrites are, based on a crude estimate of putting in less than 25% of the tube as tank water, the rest as fresh, I it looks like they are at over 20ppm. I did a 50% change last weekend and this didnt help much. I was warned by the LFS not to do too many...

I used Tetra Safe start and do not think it made a massive difference. It took over 2 weeks for Ammonia to be processed down to zero. I would consider doing it again using more scientific method for assesment, ie multiple tanks to see what worked and what didnt. Long and short of it is my tank...
