Should you remove the fish from the tank when you do water changes, in particular large water changes like 75%?
 
No, you shouldn't take the fish out.
 
Just make sure your new water is temperature matched and dechlorinated (oh, and that your heater/filter are switched off!).

Thanks I have been doing it correctly then. Can you test the water for ammonia and nitrite straight after a water change or is it best to wait

Did not know that, I've always assumed best to wait an hour before testing anything as to give tank and filter time to do their stuff to new water especially if using Seachem Prime as have heard they can give false ammonia readings if tested straight away.
 
i would do more smaller water changes rather than big ones
