But I'd rather ask and make sure I'm doing it right!!

Just about to fill my 110 gallon tank for the first time. My question is do I have to add the conditioner BEFORE it goes in the tank,or can I add it once the tank is filled??

when filling a new tank, I add the conditioner during the fill.
 
I always add it to the water in buckets before it goes in the tank as that way i can make sure it is thoroughly stirred up.
 
I do exactly the same as Tokis.
 
People with large tanks where you need a hosepipe to fill water add the conditioner to the tank first and add new water. If you use buckets you can add conditioner to each bucket.
 
If you are filling it up for the first time there are no fish in so you can add the conditioner afterwards. It is only when the tank is cycled that you need to worry about the conditioner (and obviously once you have fish in there). You've probably filled it up by now but still :p

And if it is the first time you are filling up the tank, I do not see why you would dechlorinate it since there are no fish in there and the chlorine would dissipate provided you are running your filter. Then again... if you are using old filter media.

As far as I understand the dechlorinator does not only remove chlorine, but also breaks down chloramines other harmful elements (heavy metals) from the water that will not dissipate.
 
Most tap water, from my understanding, does not have chloramines (or maybe that has been upped since my sources were written years and years ago).

I have the same size aquarium and used to use the bucket / conditioner method.

I contacted my water supply company who confirmed that they do not add Chloramine to the supply (or fluoride for that matter). What I do now is switch off the mains stopcock, wait for 4 hours for the Chlorine to dissipate, and fill the aquarium straight from the tape via a hose. The water of course, coming from the water tank.

I am a bit of a coward though, I still put water conditioner in the aquarium prior to doing this, just to be safe.

I have experienced no problems with this method and my back is a lot happier.
