Water Changes During Cycle

Do I have to put the water from the tap in a bucket first and add Prime or can't I just add the appropriate amount of Prime to the tank first and then add the water right in?  I have read so many conflicting posts I am really frustrated.  Doing a fish in cycle and I have been adding Prime to my tank and then using the Python and adding the water from the tap......Am I doing anything wrong?  I have heard that it is fine, and also heard that I have to put it all in a bucket first; which seems like a real pain.......
 
Any input is really appreciated as I want to do things correctly and just got back into the hobby after years of not having a tank.

This must have come with the prime. It is the directions right from the Seachem site;
 
Use 1 capful (5 mL) for each 200 L (50 gallons*) of new water. This removes approximately 1 mg/L ammonia, 4 mg/L chloramine, or 5 mg/L chlorine. For smaller doses, please note each cap thread is approx. 1 mL. May be added to aquarium directly, but better if added to new water first. If adding directly to aquarium, base dose on aquarium volume. Sulfur odor is normal. For exceptionally high chloramine concentrations, a double dose may be used safely. To detoxify nitrite in an emergency, up to 5 times normal dose may be used. If temperature is > 30 °C (86 °F) and chlorine or ammonia levels are low, use a half dose.

Like Twotankamin said, you can either add it to the water or add to the tank. Though, i would recommend adding it to the tank as you add the new water, not before. I'm not sure if it actually matters, but that's how I've always done it.
 
Thanks for the responses; I think I also have to just skim the top of the gravel on water changes instead of actually inserting the vacuum an inch or two in the gravel.  This was another thing I read that some people tell me to vacuum the gravel, and others say do not during the cycle.
