Co2 Valve Problems

Hi, Im kind of new to the whole CO2 thing, and was given a DIY system a few months back. But when the tank ran out obvoiusly I had to replace it this morning. When Ive come to screw the release valve on it hisses, so obviously the seal has broken, but when I unscrew the regulator valve no gas comes out. Im sure this is just something that I am doing wrong.
So any advice is much appreciated.

Basically it's a canister connected to a screw top reg valve, airline valve to a bubble counter and then an airline into a diffuser.
