Can I use a water sample from a eastabilshed tank

Just curious if I could just get a sample of water lets say a few ounces.

No, i doubt you could The bacteria doesnt just float around, it inhabits surfaces such as gravel, driftwood, etc. Try getting a used filter floss or a cupful of gravel instead.
